Currently have 2 280's in sli. I could get 1 480 now and maybe add one down the road. But would 1 be a significant difference gaming?

SSD's will make a very noticeable difference.
Pardon me but I can't comprehend the connection between comparison of these two gfx cards and liquid cooling.

one 480 no but if you plan on adding a 2nd id say yea
I went from 2 GTX 285 SLI
to 2 GTX 480 SLI
huge boost in performance because of the great scaling

If you need heavy tessellation in DX11 games right now and in the near future then you should go for GTX 480. However I would prefer waiting for the next gen if I had a pair of 280, as a single 480 won't do very impressive performance, while a pair of 480 would draw some noticeable power and dump some noticeable heat.
