Quick Vsync question

I just installed my ASUS ROG SWIFT PG279 which has 165 HZ and G sync and for one I can say I will never go back to a 60 Hz panel now

should I leave V sync on or off in the nvidia control panel (G sync in enabled)

Im running a GTX 1080
and i7 3770K
16 GB DDR3 1866 Mhz

and I play on ultra setting most of the time and my fps rarely goes above 100 in newer games like Shadow of the tomb raider runs at 70 to 90 fps for example so I just want to know the best setting for v sync
