I've used 1/2" Barbs (along with 1/2" compressions i guess) on my previous build, and I went to 3/8" because... I'm not sure anymore... I think because I needed a better bend radius.

I take it this just supports the standard mount for GPU block as shown in the picture, but can also be turned 90* so that the barbs are facing the end of the card, correct?

Unless you're looking for super low temps, no. If you want to overclock, and thus lower temps, and you wanted to be ridiculous, you could run 2 loops. But for general use, one loop is fine. ...
