measuring voltages?

I'm new to voltmodding stuff, so i was wondering: when you measure voltages you simply measure them when the system is fully hooked up and running normally? as for measuring resistance, i assume thats done when its at least turned off?

measure voltage while running, carefully!!!

measure resistance before adding components to the circuit, once they are added you cannot measure resistance!

use a digital multimeter too, because analog ones have been known to short out mobos
