What is the purpose of start button on the mobo P5Q-E ?

I have to press the start button on mobo every time I want to start my system.

How do I configure the system to start when I press the case start button?

I believe those buttons are for original set ups and overclocking uses. You should still be able to use the case power button if cables are connected correctly, just use the Q connector. On-board buttons for work benches and testing w/o the case.

its so u can check the diagnostic led and u can have it on a bench without needing the power/reset buttons
