Is there any difference between a 1/4" thread and a G-1/4" thread ?

well, it depends on what your talking about. If your on a watercooling sight, then no. If your at home depot. then yes.
G1/4" is also known as 1/4" BSPP

I have bought a D-Tek Fuzion V2 Block but want to change the 1/2" Barbs to 3/8" barbs but I was wondering because it said that the block had a G-1/4" thread size but all I could find was 1/4" thread size barbs. Will they work ok ?

Barbs require 2 sizes, the thread (which is now for the most part, all G1/4"), and the hose/tube. The latter is what that refers too.
